Overview

SS-31, also known as MTP-131, is a synthetic tetrapeptide designed for research into mitochondrial dysfunction. It carries a unique cationic 3+ charge at physiological pH, allowing efficient membrane permeability and potential passage across the blood–brain barrier. SS-31 has been widely investigated for its ability to localize in the inner mitochondrial membrane and selectively bind cardiolipin, a critical phospholipid involved in maintaining mitochondrial integrity.

Research Background

SS-31 was developed to stabilize mitochondrial function and reduce oxidative stress. Its accumulation within the inner mitochondrial membrane at concentrations reportedly exceeding plasma levels by 1000-fold suggests selective targeting. By binding to cardiolipin, SS-31 may prevent its oxidative inactivation by cytochrome c, favoring electron transport and reducing peroxidase activity. This mechanism is hypothesized to support ATP production efficiency while minimizing reactive oxygen species (ROS) leakage.
