CJC-1295 NO DAC (MOD GRF 1-29)

  • Form: Lyophilized powder
  • Purity: >99%

Overview

CJC-1295 is a synthetic peptide derivative of growth hormone–releasing hormone (GHRH), composed of 29 amino acids. It is also known as Modified GRF (1-29) or CJC-1295 without DAC. This version does not contain the Drug Affinity Complex (DAC), making it distinct from the longer-lasting DAC-modified peptide. Instead, CJC-1295 NO DAC includes four amino acid substitutions designed to improve stability and resistance to enzymatic breakdown.

Research Background

The first 29 amino acids of natural GHRH were identified in the 1980s as the minimal active sequence necessary for activity, leading to the development of Modified GRF (1-29). However, unmodified fragments suffered from rapid degradation and short half-life. By substituting the 2nd, 8th, 15th, and 27th amino acids, researchers developed CJC-1295 NO DAC as a more stable version with extended biological activity compared to the unmodified GRF (1-29).

Mechanism of Interest

  • GHRH receptor activation: CJC-1295 NO DAC binds to GHRH receptors on pituitary cells, initiating conformational changes that activate signaling pathways.
  • Signal amplification: Binding may trigger G-proteins, leading to the production of secondary messengers such as cAMP and IP3, amplifying intracellular signaling.
  • Gene regulation: cAMP-activated protein kinases may phosphorylate transcription factors, potentially influencing genes linked to growth hormone synthesis and secretion.
  • Growth hormone release: This cascade may result in vesicles containing GH merging with the pituitary cell membrane, releasing growth hormone into circulation.
